Wayrilz (rilzabrutinib) is a prescription medication approved by the U.S. Food and Drug Administration (FDA) to treat adults with persistent or chronic immune thrombocytopenia (ITP).
Wayrilz is a type of kinase inhibitor. It targets Bruton’s tyrosine kinase (BTK), a protein that helps drive certain immune signals.
Wayrilz helps change immune system activity. It can help block certain immune signals and reduce processes that lead to platelet destruction.
Doctors prescribe Wayrilz when adults with persistent or chronic ITP have had a prior ITP treatment that did not work well enough.
Wayrilz is a tablet taken by mouth. Take it at about the same times each day, with or without food.
Swallow the tablets whole with a glass of water. Do not cut, crush, or chew them.
The recommended dose of Wayrilz for ITP is 400 milligrams taken by mouth twice daily.

Sanofi, the manufacturer of Wayrilz, offers the HemAssist Copay Assistance Program. Eligible people with commercial insurance may pay as little as a $0 copay per fill. The copay program is not available to people using state- or federally funded insurance, such as Medicare, Medicaid, or TRICARE, to pay for Wayrilz.

Before starting Wayrilz, your doctor will check whether you are pregnant if you are able to become pregnant. Your doctor will also check your platelet count and liver blood tests before treatment and as needed during treatment.
Tell your doctor if you are allergic to rilzabrutinib or any ingredients in Wayrilz.
Tell your doctor about all medicines you take, including prescription drugs, over-the-counter medicines, vitamins, and herbal supplements.

Avoid grapefruit, starfruit, products that contain these fruits, and Seville oranges during treatment with Wayrilz. Seville oranges are often used in marmalades. These products may increase the amount of Wayrilz in your blood and raise your risk of side effects.
If you miss a dose of Wayrilz, take it as soon as you remember on the same day. Make sure it is at least two hours apart from your next scheduled dose.
If you are pregnant, planning to become pregnant, or breastfeeding, talk with your doctor about the risks and benefits before starting Wayrilz.
Wayrilz may harm a baby if you are pregnant. If you are able to become pregnant, use effective birth control during treatment and for one week after your last dose.
Do not breastfeed during treatment with Wayrilz and for at least one week after your last dose.

How effective is Wayrilz for ITP?
In a 24-week clinical study of adults with persistent or chronic ITP, Wayrilz helped some people have a durable platelet response. This means their platelet count stayed at a certain level for enough weeks during the study.
In the study, 23.3 percent of people taking Wayrilz had a durable platelet response. This was compared with 0 percent of people taking placebo (an inactive treatment).
The median time to first platelet response was 36 days with Wayrilz. For people taking placebo, the median time was not reached during the study.
People taking Wayrilz also needed rescue medicine less often. Rescue medicine is extra treatment used when platelet counts are too low or when bleeding needs to be managed. In the study, rescue medicine was needed by 33 percent of people taking Wayrilz and 58 percent of people taking placebo.
How long does Wayrilz take to work for ITP?
In the main clinical study, the median time to first platelet response was 36 days for people taking Wayrilz. This means that among people who had a platelet response, about half had their first response by 36 days.
Should I take Wayrilz for ITP if I have an infection?
Wayrilz can increase the risk of serious infections. These may include bacterial, viral, or fungal infections. Some infections can be life-threatening or lead to death.
In the main study, one person taking Wayrilz had fatal pneumonia. Other serious infections were also reported.
Tell your doctor right away if you have signs or symptoms of infection, such as fever, chills, or flu-like symptoms. Your doctor can decide what is safest for you.
